Conditions: Column conditioning: Polyamide CC 6 (500 g) was shaken with 3,35 l deionised water and 625 ml methanol in a 5 l-plastic container. After overnight precipitation of the polyamide the supernatant was removed, 625 ml methanol added again and the procedure repeated.
A glass column (20 x 1,4 cm i.d.) was filled with cotton wool, sea sand, swollen polyamide (high 6 cm) and sea sand. The polyamide was washed methanol-free with 100 ml deionised water.
Sample application: The concentrated extract was applied to the top of the column.
Column washing: The flask was rinsed twice with 5 ml deionised water in total, and then the column was washed with 25 ml water.
Elution: The polyphenols were eluted in three steps: 30 ml methanol (fraction M1), 90 ml methanol (fraction M2) and 40 ml DMF (fraction D)(approximately 60 drops/min).
Evaporation: Fraction M1 and fraction M2 were concentrated to about 1 ml, and fraction D evaporated to dryness. All residues were transferred to a 5 ml flask with acetic acid (2%):acetonitrile (98:2, v/v), filled up with the same solution and filtered through a 0,2 μm PTFE filter (polytetrafluorethylene).
